High-quality labeled antibodies are crucial for many research applications including Western blotting, ELISA, immunofluorescence, immunohistochemistry, and flow cytometry. Specialized conjugates are not always commercially available but can be custom produced to your specifications at Proteos.

Custom labeling (conjugation) of monoclonal antibodies can be included with our antibody production services.  Click here for more information regarding recombinant antibody production in HEK293 and CHO cells.  We have extensive experience with purification of full length antibodies and antibody fragments (Fab, (Fab’)2, scFv) from cell culture supernatant or customer provided ascites covering a variety of host species.  Labeling can also be requested for customer provided purified antibodies or antibody fragments.

Proteos offers a variety of labels for conjugation to monoclonal antibodies. Examples of labels include:

  • Fluorescent molecules (FITC, rhodamine, Alexa Fluor® dyes, etc.)
  • Small molecules (biotin)
  • Enzymes (HRP or AP)

The degree of labeling on antibodies can have a large impact on downstream applications. We offer a full suite of protein analytics to help determine and optimize the molar ratio of label to antibody. Available analytics include spectrophotometric measurements, HPLC, plate based assays, SDS-PAGE, mass spectrometry, and flow cytometry. We also offer thermal shift assays to assess antibody stability and ligand binding.

For many commonly used antibody labels, a simple calculation based on spectrophotometric readings can determine the degree of labeling. An example of calculating the degree of labeling for an antibody with rhodamine is shown to the right. This is a simple method requiring only 2 µL of sample and robust enough for most applications.
